- ItemEtiology and epidemiology of digital dermatitis in Australian dairy herds.(Elsevier B.V., 2024-02-06) McPherson A; Tranter B; Phipps A; Laven R; House J; Zadoks RN; Rowe SABSTRACT Bovine digital dermatitis (BDD) is an important cause of lameness in dairy cows worldwide. However, there is very little known about this disease in Australian herds, which are predominantly managed on pasture. The primary objectives of this cross-sectional study were to describe the presence and prevalence of BDD in Australian dairy herds and to characterize the microbiota of healthy skin and M4 lesions of BDD-affected, pasture-managed cows. Cows from 71 dairy herds were examined at milking time to identify the presence of BDD lesions. True prevalence was estimated using Bayesian methods with informative priors for sensitivity and specificity. Biopsy samples (n = 60) were collected from cows with and without BDD-lesions in 7 pasture-based herds. The microbiota in the superficial and deep strata of each tissue biopsy were characterized via sequencing of the V3-V4 region of the bacterial ribosomal RNA gene. Lesions were detected in 1,817 (11.5%) of 15,813 cows, and in 68 of 71 (95.8%) herds. The median herd-level apparent and true prevalences of BDD were 8.5 and 18.1%, respectively, but this varied considerably between farms. On farms with BDD, M4 lesions accounted for 70 to 100% of all lesions (interquartile range = 95.1 to 100%, median = 100%). M2 lesions (i.e., large ulcerative lesions) were observed at low prevalence (<2.2%) in the few herds (7/71, 9.9%) where they were found. There was a statistically significant difference in the composition of the microbiota between healthy skin and M4 lesions but not between superficial and deep tissue layers. Several gut- and effluent-associated bacterial taxa, including Lentimicrobium and Porphyromonas, which have previously been associated with BDD, were abundant in BDD lesions but not in control biopsies. Our study supports the idea that such taxa are involved in, though possibly not essential to, lesion development and persistence in pasture-managed cows in Australia. Our results also suggest that Dichelobacter may contribute to the disease process. We conclude that BDD is likely to occur in most Australian dairy farms, but that further studies are needed to identify its impact on cow welfare and productivity. Further investigation of the etiology of BDD in Australian dairy herds is also necessary to inform prevention and control strategies. INTERPRETIVE SUMMARY: We visited 71 farms in Australia to estimate the prevalence of bovine digital dermatitis (an infectious skin disease that causes lameness in cows). Most farms (68 / 71) were identified to be endemic for the disease, but that the painful lesion types (score = M2) were uncommon. Microbiome analysis was conducted on 60 biopsy sample of cows with and without BDD lesions. We found that cows with BDD had higher abundances of gut- and effluent-associated bacterial taxa, including Lentimicrobium and Porphyromonas. We conclude that BDD is present on most Australian dairy farms.
- ItemFood Safety, Health Management, and Biosecurity Characteristics of Poultry Farms in Arusha City, Northern Tanzania, Along a Gradient of Intensification(East African Health Research Commission of the East African Community, 2019-05-24) Sindiyo E; Maganga R; Thomas KM; Benschop J; Swai E; Shirima G; Zadoks RNBACKGROUND: With the growth, urbanisation, and changing consumption patterns of Tanzania's human population, new livestock production systems are emerging. Intensification of poultry production may result in opportunities and threats for food safety, such as improved awareness of biosecurity or increasing prevalence of foodborne pathogens including nontyphoidal Salmonella or Campylobacter spp. We conducted a semiquantitative analysis of poultry production systems in northern Tanzania, with emphasis on biosecurity, health management practices, and prevalence of foodborne pathogens, to gain insight into potential associations between intensification and food safety. METHODS: Interviews were conducted with managers of 40 poultry farms, with equal representation of 4 production systems (extensive, semi-intensive, or intensive production with indigenous chickens, and broiler farming). Per farm, up to 10 birds (total, 386) were tested for cloacal shedding of nontyphoidal Salmonella, with a subset of farms tested for Campylobacter. Data were analysed using univariate statistics, and results were discussed during feedback workshops with participating farmers and extension officers. RESULTS: Clear differences existed between farm types with regard to implementation of biosecurity and health management practices and use of extension services. By contrast, prevalence of foodborne pathogens (6 of 40 farms or 15% for nontyphoidal Salmonella and 13 of 26 farms or 50% for Campylobacter spp.) was not farm-type specific, indicating that it is driven by other factors. Across farming systems, knowledge and awareness of the presence of antimicrobials in poultry feed and the need to abide by post-treatment withdrawal times were limited, as was access to impartial professional advice regarding treatment. CONCLUSION: Different control measures may be needed to protect poultry health compared to public health, and improvements in information provision may be needed for both.

Africa is estimated to have the world's highest incidence of campylobacteriosis and a relatively high prevalence of AMR in Campylobacter spp. from humans and animals. Few studies have compared Campylobacter spp. isolated from humans and poultry in Africa using whole-genome sequencing and antimicrobial susceptibility testing. We explored the population structure and AMR of 178 Campylobacter isolates from East Africa, 81 from patients with diarrhea in Kenya and 97 from 56 poultry samples in Tanzania, collected during 2006-2017. Sequence type diversity was high in both poultry and human isolates, with some sequence types in common. The estimated prevalence of multidrug resistance, defined as resistance to >3 antimicrobial classes, was higher in poultry isolates (40.9%, 95% credible interval 23.6%-59.4%) than in human isolates (2.5%, 95% credible interval 0.3%-6.8%), underlining the importance of antimicrobial stewardship in livestock systems.
- ItemSpread of Nontyphoidal Salmonella in the Beef Supply Chain in Northern Tanzania: Sensitivity in a Probabilistic Model Integrating Microbiological Data and Data from Stakeholder Interviews(Wiley Periodicals LLC on behalf of Society for Risk Analysis, 2022-05) Zadoks RN; Barker GC; Benschop J; Allan KJ; Chaters G; Cleaveland S; Crump JA; Davis MA; Mmbaga BT; Prinsen G; Thomas KM; Waldman L; French NPEast Africa is a hotspot for foodborne diseases, including infection by nontyphoidal Salmonella (NTS), a zoonotic pathogen that may originate from livestock. Urbanization and increased demand for animal protein drive intensification of livestock production and food processing, creating risks and opportunities for food safety. We built a probabilistic mathematical model, informed by prior beliefs and dedicated stakeholder interviews and microbiological research, to describe sources and prevalence of NTS along the beef supply chain in Moshi, Tanzania. The supply chain was conceptualized using a bow tie model, with terminal livestock markets as pinch point, and a forked pathway postmarket to compare traditional and emerging supply chains. NTS was detected in 36 (7.7%) of 467 samples throughout the supply chain. After combining prior belief and observational data, marginal estimates of true NTS prevalence were 4% in feces of cattle entering the beef supply and 20% in raw meat at butcheries. Based on our model and sensitivity analyses, true NTS prevalence was not significantly different between supply chains. Environmental contamination, associated with butchers and vendors, was estimated to be the most likely source of NTS in meat for human consumption. The model provides a framework for assessing the origin and propagation of NTS along meat supply chains. It can be used to inform decision making when economic factors cause changes in beef production and consumption, such as where to target interventions to reduce risks to consumers. Through sensitivity and value of information analyses, the model also helps to prioritize investment in additional research.
